   <subfield code="a">Bixin is a carotenoid pigment found in the outer layer seeds of B. orellana found to display multiple biological activities, some of the more prominent ones being its anti-inflammatory and antioxidant properties. Even with these promising potentials, formulating with Bixin has posed to be challenging due to its issues with solubility, and the stability of its nanoformulation in the dispersion system. To overcome these problems, formulations of bixin-loaded chitosan-tripolyphosphate hyaluronic acid-coated nanoparticles (HA-Bx-CSS/TPP NPs) have been produced using ionic gelation to improve its stability in the dispersion system preventing aggregation. Factors such as Bixin Concentration, Chitosan:Tripolyphosphate ratio (CS:TPP), and Hyaluronic Acid (HA) concentration have been explored to determine their effects on the characteristics of the HA-Bx-CSS/TPP nanoparticles through the analysis of Particle Size, Zeta Potential, and Encapsulation Efficiency. Preliminary tests have also been conducted to obtain the maximum solubility of 40% Bixin which is 12mg/mL, and set the appropriate levels for each factor. Each factor was set with five (5) different levels, and each level was used in the formulation. It was found that the Bixin levels three (3) and four (4), with 9 mg/mL and 10 mg/mL , CS:TPP levels (3) 4:1 and (4) 4.25:1, HA levels (2) 0.3% and (3) 0.5% are the best levels for formulation since they resulted in a nanoparticle suspension with no aggregation, &lt;1000 nm size, acceptable PDI, and zeta potential ≥ +/-30mV. Further optimization has been performed and it was found that concentrations of Bixin 9 mg/mL and 10.5 mg/mL in combination with CS:TPP 4:1 and HA 0.3% are the best formulations with %EE 45.54% and 45.20%, respectively.
